چکیده

doi: 10.1111/j.1399-3054.2006.00739.x To investigate selenium tolerance mechanisms in Arabidopsis thaliana, genetic and physiologic studies were performed in the three Arabidopsis accessions Landsberg erecta (Ler), Columbia (Col) and Wassilewskija (Ws). Accession Ler was significantly less tolerant to selenate than Ws and Col, whereas Ws was less tolerant to selenite than the others. Analysis of selenium tolerance in F1 and F2 plants obtained from crosses between these accessions suggest that multiple genes are involved in selenate tolerance and a single major gene controls selenite tolerance in these populations. Bulked segregant analysis in two F2 populations indicated that molecular marker ciw7 on chromosome 4 is linked to selenite tolerance, and three molecular markers on chromosomes 1, 3 and 5 (nga111, ciw4 and ciw8, respectively) are linked to selenate tolerance. The ecotypic variation in selenite tolerance appeared to be correlated with root levels of non-protein thiols. Also, the shoot tissue levels of selenocysteine (SeCys) and selenocystine were correlated with tolerance to both selenate and selenite. Judging from RT-PCR results, several sulfate transporters and S assimilatory enzymes appear to be upregulated by selenate and selenite at the transcriptional level. A potential SeCys methyltransferase was expressed at lower levels in selenite-sensitive Ws than in the other two, when grown on selenite. Together, these studies show that there is substantial intraspecific variation in tolerance to selenate and selenite in Arabidopsis, and provide insights into the genetic and biochemical mechanisms underlying the observed ecotypic differences.

Variation in selenium tolerance and accumulation among 19 Arabidopsis thaliana accessions.

Selenium (Se) is an essential element for many organisms but also toxic at higher levels. The objective of this study was to identify accessions from the model species Arabidopsis thaliana that differ in Se tolerance and accumulation. Nineteen Arabidopsis accessions were grown from seed on agar medium with or without selenate (50 microM) or selenite (20 microM), followed by analysis of Se toler...

Gene transcriptomic profile in arabidopsis thaliana mediated by radiation-induced bystander effects

Background: The in vivo radiation-induced bystander effects (RIBE) at the developmental, genetic, and epigenetic levels have been well demonstrated using model plant Arabidopsis thaliana (A. thaliana). However, the mechanisms underlying RIBE in plants are not clear, especially lacking a comprehensive knowledge about the genes and biological pathways involved in the RIBE in plants. Materials and...

Yeast Two Hybrid cDNA Screening of Arabidopsis thaliana for SETH4 Protein Interaction

SETH4 coding sequence with 2013 bp is a member of gene family expressed in gametophytic tissues of Arabidopsis thaliana. This fragment was PCR amplified using Kod Hi Fi DNA polymerase enzyme. This fragment was cloned into pGBKT7 bate vector and transformed E. coli DH5? cells containing vector were selected on LB medium containing Kanamycin. Finally, pGBKT7-SETH4 bate was transformed into yeast ...

Mapping quantitative trait loci associated with selenate tolerance in Arabidopsis thaliana.

Selenium is essential for many organisms, but is toxic at higher levels. To investigate the genetic basis of selenate tolerance in Arabidopsis thaliana, quantitative trait loci (QTL) associated with selenate tolerance in accessions Landsberg erecta and Columbia were mapped using recombinant inbred lines (RILs). The selenate tolerance index (TI(D10) = root growth + 30 microm selenate/root growth...

Negative control of Strictisidine synthase like-7 gene on salt stress resistance in Arabidopsis thaliana

Strictosidine synthase-like (SSL) is a group of gene families in the Arabidopsis genome, which whose orthologues in other plants are key enzymes in mono-terpenoid indole-alkaloid biosynthesis pathway. The SSL7 is upregulated upon treatments of Arabidopsis plants with signaling molecules such as SA, methyl jasmonate and ethylene. To find the functional role of the gene, a T-DNA-mediated knockout...
